Wycombe, dramatic 3-2 winners at Northampton on the final day of the season, finished 13 points ahead of Plymouth over the course of the campaign while Southend were 12 ahead of Stevenage.
But the play-offs offer all four teams a shot at a Wembley final and promotion to League One and the results will be difficult to predict with Wycombe manager Gareth Ainsworth predicting an 'intense' atmosphere at Plymouth's Home Park on Saturday night.
The play-offs are Northampton's aim as a minimum next season with manager Chris Wilder starting his preparations earlier this week with the release of nine players.
Speaking about his recruitment plans to the Chronicle & Echo, Wilder said: “I am not a lover of short-term loans, and if we are going to sign anybody on loan it will be a season-to-season agreement, or a windwow-to-window, which will mean August to January."
